The challenge will see walkers cover 100 miles in 48 hours with no nights of resting.
Some 500 walkers, and a handful of runners, will start at Wadebridge and head towards Teignmouth, passing through various checkpoints, including a breakfast stop in Tavistock.
Although sections will be on minor roads, most of the route will be on public footpaths which, given the recent weather conditions, has been dubbed 'a truly grim challenge'.
A variety of walkers will be taking part from fit athletes to pensioners. The walk takes place over the bank holiday weekend, May 25 to 27.
